Abstract A comprehensive design chemical experiment of synthesis of 5-(4-antipyrine azo) salicylaldehyde and determination of niobium(Ⅴ)by spectrophotometric method and its teaching plan were introduced. The homemade antipyrine azo salicylaldehyde was used as chromogenic reagent in color reaction with niobium (Ⅴ) ion. The effect of sensitization of the color reaction about 5-(4-antipyrine azo) salicylaldehyde and niobium (Ⅴ) in the presence of pairing surfactant compound was discussed. Through this comprehensive design chemical experiment, the students' understanding and applying capability of some certain knowledge about organic synthesis, physical chemistry and analytical chemistry were enhanced, the students' practical ability and the comprehensive innovative ability were improved. It is beneficial for training practical talents.
