Abstract This paper introduced the stratified progressive teaching design of secondary structure of nucleic acid as following:the interaction force leading to the secondary structure of nucleic acid as the core, inspiring students to think about the possible interaction patterns between nucleic acid bases under the influence of hydrogen bond and π-π stacking interaction, introducing the structural polymorphism of nucleic acid, then extending to the secondary structures of nucleic acid. This teaching process could fully stimulate students' learning interests and divergent thinking ability, which would be helpful for students to understand the secondary structure of nucleic acid.
