Abstract A comprehensive experiment has been designed named determination of tocopherols in cereal grains by using dispersive liquid-liquid microextraction (DLLME) coupled with high performance liquid chromatography(HPLC), and was applied to the chemical basic comprehensive experiment teaching. This experiment is designed on the basis of DLLME, a novel sample pretreatment technique. By consulting the relevant literature, the suitable extraction mode, dispersion strategy, extraction device, evaluating indicators of extraction effect, separation and analysis conditions of HPLC were determined. The design is based on recent scientific research, which can strengthen the operational skills of basic chemistry experiments, and arouse the students' enthusiasm in exploring the unknown and enhance the innovative ability and overall quality.
