Abstract Based on the importance of the experiment of separating amino acids by paper chromatography in undergraduate specialties such as biology and medicine, this paper makes a detailed supplement to the part of the experimental textbook which can not be explained in detail, including the stationary phase of paper chromatography, the capillary effect of filter paper, the role of water and acid (base) in mobile phase. It will deepen students' understanding of the principle of paper chromatography.
