Abstract This paper outlined the application of problem-oriented teaching mode in the chapter on organic spectrum analysis in the course of medical organic chemistry for medical colleges. This part has the "three difficult" teaching characteristics of the students "difficult to learn" and "difficult to test" and the teacher "difficult to teach". We conducted theoretical teaching of the core concept of organic spectrum analysis (introduction) through layer-by-layer questions, sequential method of teaching, and rich application examples. It can effectively improve students' interest in learning, reduce the difficulty of learning, and enable students to master the basic theory and core concepts of organic spectrum analysis in a limited number of hours, and lay a solid foundation for the cultivation of subsequent spectrum dissociation ability.
