Abstract The paper described an instructional design of deprotection of amino group through open experimental design. The students were encouraged to design and carry out the orthogonal experiments of acetanilide hydrolysis under acidic or basic conditions in groups, using recycled acetanilide as the starting material to prepare aniline as the product. The optimal condition and procedure of the experiments were confirmed by comparing the yields of the hydrolysis under different conditions. The results of the experiments conducted by students in the past 3 years were summarized and analyzed, and the repeatability of the results was investigated by verification experiments.
