Abstract During the construction of “double first-class” universities, it is important to promote the cross integration of theoretical disciplines and practical disciplines. Under this background, this paper takes the “magical methyl effect” as an example to illustrates the fusion of the courses of medical chemistry and pharmaceutical chemistry. The influence of the magic methyl group on the pharmacokinetic properties, molecular conformation or lipophilicity of drugs was analyzed from their chemical structures, which finally eliminates their toxicity, energetically favors binding and boosts potency, or increases their druggability. The integration of the comprehensive and practical course of pharmaceutical chemistry and the theoretical course of medical chemistry will help students understanding the classical theories of chemistry and sets up a meaningful knowledge system based on the reality of new drug-development that is meaningful for students’ career growth. It is hoped that the shortcoming of the disconnection of theoretical theories and practical applications in the traditional teaching modes can be overcome, and the standards of education could be constantly improved.
