Improvements and Teaching Discussions of Thin-layer Chromatography Experiment

Abstract  A mixture of N,N-dimethyl-4-(phenylazo)benzenenamine and trans azobenzene was selected as a test sample, which was expanded in single and mixed solvent with different ratio after being spotted on a TLC plate. The influences of sample polarity and solvent polarity on the separation effects of the thin-layer chromatography have been investigated. Acetanilide and benzamide were selected as test samples, which were developed under the ultraviolet lights and in iodine vapor after being expanded.Via examples above, students could master common chromogenic methods in the thin-layer chromatography experiments. A few problems in processes of experiments, such as selections of matter polarity and developing solvents, different chromogenic methods and separation effects were discussed on teaching methods.
