Abstract In order to cultivate students' ability in preparing comprehensive experiments as well as promote the greenization and standardization of chemistry teaching experiments, we have improved the original experiment on synthesizing cyclodextrin derivatives by changing the reaction medium from pyridine to NaOH aqueous solution, and designedthe preparation experiment oftwo 6-mono-substituted β-cyclodextrin derivatives. This experiment used β-cyclodextrin as a raw material to react with p-toluenesulfonyl chloride to produce mono-(6-O-p-toluenesulfonyl)-β-cyclodextrin, further reacted with ethylenediamine to prepare mono-(6-ethylenediamine-6-deoxygen)-β-cyclodextrin. The experiment involves operations such as filtration, recrystallization, and the products are characterized by infrared spectroscopy, nuclear magnetic resonance hydrogen spectroscopy, melting point determination as well as high-performance liquid chromatography. This comprehensive design experiment achieves the integration of knowledge in organic chemistry and analytical chemistry, which can help students who have the need of scientific research or want to engage in relevant work lay a solid experimental foundation.
