Abstract  This paper introduced an original application of determination of high polar and hydrophilic compounds by high performance liquid chromatography (HPLC), to extend the HPLC application scopes of the instrumental analysis textbook and inspire students comprehensively understand characteristics of HPLC technology. Merck ZIC®-HILIC column was used to perform chromatographic separation and establish the quality control method for the typical medical and chemical product, cytosine. This paper introduced hydrophilic interaction liquid chromatography(HILIC), experiment parameter optimization and detailed methodology validation in order to make the method legibly understood by readers and increase a new teaching case for instrumental theory and experiment class in life science, pharmaceutical and chemical majors.
